I don't know about ALL dogs, but in greyhound of racing stock (AKC greyhounds are a totally different animal) there is no doubt in anyone's mind that "spookiness" is an inherited chemical imbalance. A spooky greyhound is one who, when the same trainer approaches their crate every day, the dog looks and acts as though she has never seen the person before. They are afraid of people, dogs, shadows. It is rare that behavior modification makes a difference when used alone. Any breeder that has had a spook in their lines will tell you that spooky moms have spooky pups. A few have more mild symptoms and have successful careers and are used for breeding. Most are never bred and some are not even suitable as pets, as they become fear biters and are then euthanized. As far as dogs in general, the heritability of behavior issues is still up for study.
